内容記述 | We studied the effects of 10 drugs known to be inhibitors of ornithine decarboxylase (ODC), S-adenosyl-methionine decarboxylase (SAMDC), spermidine (Spd) synthase or spermine (Spm) synthase on the polyamine synthesis of the budding yeast Saccharomyces cerevisiae and the fission yeast Shizosaccharomyces pombe. Cellular polyamines were analyzed by high-performance liquid chromatography after culture in the absence or presence of the inhibitors. α-Difluoromethylornithine (DFMO), methylacetylenicputrescine (MAP) and α-hydrazinoornithine (HOrn), at 1-10mM, lowered putrescine (Put) levels in both yeasts, showing inhibition of the ODC activity. Trans-1,4-diamino-2-butene (DAB) at a concentration of 5mM decreased Put levels. The decrease in Spd and Spm levels suggests inhibition not only of ODC activity but also of Spd synthase and Spm synthase activities by DAB. At 5-10mM, 2-mercapto-ethylamine (MEA) and 3-methylthiopropylamine (MTPA) decreased Spd and Spm levels in each yeast. Put levels were rather increased by these two drugs, although the magnitude of Put increase was not the same between S. cerevisiae and S. pombe. Methylglyoxal-bis (guanylhydrazone) (MGBG), at 1-5mM, decreased Spd and Spm levels, suggesting inhibition of SAMDC activity as in mammalian and protozoan cells. Addition of 5-10mM of cyclohexylamine (CHA), 4-methylcyclohexylamine (MCHA) or N-(3-aminopropyl)cyclohexylamine (APCHA) resulted in decreases in cellular Spd and Spm levels, of both yeasts. CHA and MCHA appeared to preferentially inhibit Spd synthase activity, while APCHA specifically supressed Spm synthase activity. An increase in Put levels induced by the three drugs was observed in S. pombe but not in S. cerevisiae. It was suggested that the inhibitors are significantly effective against the polyamine synthesis of the yeasts, S. cerevisiae and S. pombe, as in mammalian and protozoan cells. However, there are some differences in sensitivity of these yeasts to the inhibitors tested. | |||||||||
