| 内容記述 | A smear specimen is usually dried by blowing cool air. But we are not exactly sure why we use cool air for drying smears. We studied the effects of drying temperature upon the morphological change of blood cells in both the Romanowsky stain (Wright method) and the cytochemical peroxidase reaction. In the Wright stain, the rate of hemolysed red blood cells increased in proportion to the rise in the drying temperature of the smear. However, the morphological state of the white blood cells remained unchanged. In the cytochemical peroxidase reaction, myeloperoxidase (MPO) positive granules of neutrophil leukocytes and monocytes decreased in proportion to the rise in the drying temperature of the smear. And then the neutrophil MPO score significantly decreased depending on the drying temperature. | |||||
